I guess this would go for any randomizer file, but basically there's a program for LttP that takes a Japanese 1.0 ROM and creates a new .sfc that randomizes chest contents.
Is there a way to create a .sfc file and turn it into a patch that can be applied to a USA version of LttP?

Yeah it's possible. Should be pretty easy really using 3 things: A US rom of Link to the Past with the header removed, the output sfc that you're referring to, and the program lunar ips. Lunar ips will create a patch using the roms I spoke of (using US Lttp as your base rom and the new rom with randomized chests as the end result)
